リンパ腫瘍におけるサイクリンD1トランスクリプトbの役割と診断、治療への応用

研究概要

我々が独自に単離したサイクリンD1トランスクリプトbのリンパ腫瘍における役割ならびに診断治療への応用をめざし、今年は以下の諸点を明らかにした。
1.従来のサイクリンD1(トランスクリプトa)を過剰発現している固形癌において、サイクリンD1トランスクリプトbの発現は極めて低いが、t(11;14)転座を示すリンパ腫瘍細胞株でサイクリンD1トランスクリプトbが特異的に高発現していることを見い出した。次に、lac operonを用いて、マウスBAF3pro-B細胞でサイクリンD1トランスクリプトa,bを自由に発現誘導する系を樹立した。両トランスクリプトa,bを同時に認識するモノクローナル抗体を用いて、初めて細胞内でトランスクリプトb産物を同定した。
2.実際のリンパ腫患者検体を検索し、マントル細胞性リンパ腫で特異的にサイクリンD1トランスクリプトbが高発現していることを見い出した。
3.サイクリンD1,D2,D3の発現を各構造血器腫瘍細胞株と患者検体を検索して、リンパ球系ではそれぞれのサイクリンが特異的な機能を有すること、骨髄球系ではこれらのサイクリンの発現に特徴的な差はないことが明らかとなった。
