分裂酵母における減数分裂特異的な組み換え機構の解析

研究概要	差分化cDNAライブラリーを作成することで、これまでに分裂酵母の減数分裂特異的に転写誘導される遺伝子を現在までに27個単離した。この一つeta1^+(enhanced transcript after nitrogen starvation)遺伝子はノーザン解析をすると1.0kbと2.8kbの2本のバンドを示し、このうち2.8kbのバンドのみが窒素源枯渇後数時間後に劇的に転写誘導されるという減数分裂特異性を示した。両方に対応するcDNAを単離して塩基配列決定すると両方ともポリA付加の位置は同一で共通にRad24の読みとり枠(ORF)を含むが、後者は余分な5'上流(1.8kb)にもうひとつの大きなORFを含むことが分かった。このORFは遺伝子組み換えに関わる出芽酵母のDmc1(大腸菌RecAのホモログ)と酷似していたので、コードする遺伝子をdmc1^+と名付け機能解析した。これらがbicistronic mRNAとし転写されている事実はプローブを変えてノーザン解析やゲノムサザン解析を行うことで再確認した。3種類の遺伝子破壊株(dmc1Δ,rad24Δ,dmc1Δrad24Δ)の解析から、いずれの遺伝子の破壊も減数分裂には何ら影響を与えないことが分かった。この結果は出芽酵母のDMC1が減数分裂(とくに組み換え)に必須であることに比べて特異である。ただし、テトラド解析によるとdmc1破壊株は減数分裂過程でに組み換えに異常を示し、遺伝子転換効率も野生株に比べて顕著に低下していたことは分裂酵母のDMC1もやはり遺伝子組み換えに関わっていることを示唆する。昨年報告したMeu13の機能と考え合わせると我々の方法は減数分裂組み換えに関わる遺伝子の包括的単離にも有用であることが分かった。
