新規抗マラリア剤アルテミシニン及びアルカロイド類縁体の完全合成と作用機序の解明

研究概要	新規抗マラリア剤の開発を目的として研究を行い、以下の成果を挙げることができた。 1.アルテミシニン類縁体の合成研究:クロロキン耐性マラリア原虫に対して、強い活性を示すセスキテルペンの一種であるアルテミシニンの類縁体の合成を目的として研究を行なった。その結果分子内ディールス・アルダー反応によって合成したデカリン誘導体から、アルテミシニン類縁体の合成に成功した。合成品は10^<-8>モル濃度で熱帯熱マラリア原虫の成育を阻止し、1000倍以上の選択毒性を示すことが確認された。本合成ルートの改良、反応成績体の構造決定、他類似化合物の合成をさらに検討している。 2.ロダシニアン系色素の抗マラリア活性:脂溶性非局在化カチオン性分子はエネルギー生産を活発に行うミトコンドリアに集積すると言われている。そこで種々の色素について抗マラリア活性を検討したところ、ロダシアニン系色素中に10^<-8>モルオーダーで熱帯熱マラリア原虫の成育を抑制し、214倍の選択毒性を示す化合物が発見された。現在リスザルを用いてのin vivo試験を行なっている。なお本化合物は熱帯熱マラリア原虫の組織を選択的に蛍光染色することが確認され、生化学的ツールとしての利用が期待されている。
