マラリア感染に対する防御免疫機構-赤内型P.berghel強毒株NK65とその弱毒株XATを用いた解析-

研究概要	昨年度までの検討により、IL-12およびIFN-γ産生とその下流に位置する感染防御のEffector機構として脾臓のマクロファージによる貪食能が感染赤血球の除去に重要であることを明らかにした。そこで、今年度は、さらに抗体依存的なFcレセプター(R)を介した貪食およびADCC(Antibody-Dependent Cell-Mediated Cytotoxicity)活性を消失しているFcRγ鎖のノックアウトマウス(KO)とIFN-γKOマウスを用い検討を行った。まず、弱毒株感染後、脾細胞の感染赤血球に対する貪食能を抗XAT抗血清存在下および非存在下で経時的に測定すると感染赤血球投与後1週間後頃から抗体依存的な貪食能が上昇しピークに達しその後徐々に減少することがわかった。次に、FcRγ鎖KOマウス由来の感染脾細胞を用い同様に貪食能を測定すると、抗血清存在下でも抗体依存的な貪食能は見られず、さらにIFN-γKOマウス由来の感染脾細胞を同様に用いても貪食能はコントロールのWild-Typeマウスに比べ有意に低下していることがわかった。そこで、次にこれらのマウスの弱毒株感染に対する抵抗性を調べると、FcRγ鎖KOマウスでは抵抗性が減少しほとんどのマウスで第2ピーク後Parasitemiaが上がり続け死亡した。この時感染脾細胞からのIFN-γ産生や血清中の抗XAT抗体価はコントロールのWild-Typeマウスとほぼ変わらなかった。さらに、IFN-γ KOマウスでは全てのマウスが感染赤血球を除去できず死亡したが、この際感染脾細胞の抗体依存的な貪食能および血清中の抗XAT抗体価も有意に低下しており、これらの機能低下が感染防御能の消失につながったと考えられる。.以上より、IFN-γ産生の下流に位置するEffector機構としてFcRを介した抗体依存的な貪食能の重要性が明らかになった。
