ヒトT細胞白血病ウイルスによる自己免疫発症機構の解析

研究概要

我々は先にHTLV-Iのtax遺伝子を導入したトランスジェニックマウス(HTLV-I-Tgマウス)を作製し、このウイルスがヒト関節リウマチに似た慢性関節炎を引き起こすことを示した。また関節炎の発症には、T細胞自己免疫およびIL-1、IL-6の過剰産生が関与することを明らかにした。本年度はIL-1の病態形成への関与の仕方を明らかにするために、IL-1およびIL-1レセプターアンタゴニスト(IL-lra)欠損マウスを用いて、免疫系に対する影響を検討した。
1.HTLV-I-TgマウスのIL-1を欠損させると、関節炎の発症率が約半分に低下した。このマウスでII型コラーゲンに対する抗体産生、T細胞の反応性を検討し、応答が低下していることがわかった。
2.IL-1 KOマウスにLPSを投与したところ、MIP-1αやL-セレクチンの誘導が弱かった。
3.IL-1 KOマウスを羊赤血球で免疫したところ、抗体産生能が低下しており、IL-1は抗体産生に重要な役割を果たしていることがわかった。これらのマウスのT、B、APCの増殖能、抗原提示能は正常であったが、T細胞とAPCの相互作用がうまく機能していないことがわかった。
4.IL-1ra KOマウスは、多発性の慢性関節炎を発症した。このマウスでは、リウマチ因子などの血中レベルが亢進しており、自己免疫になっていることがわかった。このことは、IL-1raの異常に基づく自己免疫が、関節炎の原因であることを示唆している。
以上の結果から、HTLV-I-Tgマウスでは、関節で過剰産生されたIL-1により、種々の接着因子やケモカインの産生、あるいはT細胞とAPCの相互作用が活性化される結果、免疫系が過剰に活性化され、自己免疫になることが示唆された。
