IL-クレセプターによるTCRγ遺伝子のV-J組み換え誘導機構

研究概要

インターロイキン7(IL-7)はリンパ球前駆細胞の増殖因子であり、生体内におけるリンパ球の分化にきわめて大きな役割を果たしている。本研究の目的は、IL-7レセプター(IL-7R)のシグナル伝達分子のSTAT5によるgermline転写の誘導が、TCRγ遺伝子座のDNA組換え酵素に対するaccessibilityを上昇させV-J組換えを誘導する機構を解明することにある。
まず、IL-7R欠損マウスの胎児肝臓のT前駆細胞にレトロウイルスを用いてIL-7RやSTAT5を導入し、胎児胸腺器官培養でT細胞に分化させた。活性型STAT5を導入したものでは、IL-7R導入時ほどではないが細胞数がある程度回復し、γδT細胞の分化が見られた。したがって、活性型STAT5の導入がIL-7R欠損マウスにおけるγδT細胞の分化を回復させたことが示された。
次に、STAT5が転写共役因子であるCBPやp300と協調して転写を活性化するかを調べた。転写共役因子は内在性のヒストン・アセチル化酵素活性を持ち、周辺のヒストンをアセチル化しヌクレオソーム構造をゆるめて、転写装置が働くことを促している。5'Jγ1プロモーター領域をBa/F3細胞に導入すると、STAT5依存的およびCBP/p300依存的に転写が活性化された。さらに、Jγ1領域のヒストンが高アセチル化されていることが、染色体免疫沈降法により示された。
次に、STAT5によりTCRγ遺伝子のgermline転写が誘導されている時に、RAG1とRAG2が存在するとDNA切断がおこるかを解析した。Ba/F3細胞にRAG1と2の発現ベクターを導入すると、Jγ遺伝子は高頻度に切断されたがIg遺伝子やTCRβ遺伝子は切断されなかった。この結果は、転写装置にとってaccessibleな状態はDNA組換え酵素にとっても同じであることを示している。
