T細胞系幹細胞の集積する新しい腸管リンパ組織の研究

研究概要	マウス腸管粘膜固有層に約1,000個の未分化T細胞の集積するリンパ組織が多数存在することを見出し、これらをcryptopatches (CP) と命名した。ヒトやマウスの腸管上皮細胞間にはintestinal intraepithelial T lymphocytes (IEL) と呼ばれる腸管独特のT細胞が分布し、全末梢T細胞の1/2〜1/3を占めるが、その生体内生理的機能は良く分かっていない。我々はCPがIEL前駆細胞の発達分化局所であることを証明したが、これに引続き、CPの形成機序やCP内でのIEL前駆細胞の発達分化機構を解明することは、IELの生体内生理的機能追及に必須と考えられた。CPの組織形成に重要なサイトカインとしてIL-7とIL-15が関与することを明らかにするとともにIL-2Rγ鎖ミュータント(γ^<-/->)マウスの腸管粘膜固有層のCPは検出限界以下であることも確かめられた。Γ ^<-/->マウスでは、IL-2R、IL-4R、IL-7R、IL-9R及びIL-15Rからのシグナル伝達が傷害されるが、CP形成不全はIL-7RとIL-15R機能の双方が傷害されることによると考えられる。次に胸腺及びCPを欠如するnu/nuγ^<-/->マウスと胸腺を欠如するもののCPは十分存在するnu/nu SCID マウスに存在するT細胞レセプター(TCR)を発現しないIELを細胞及び細胞下レベルで比較追求した結果、IEL前駆細胞がCP内で発達分化する過程でIEL特有のCD8αα分子やαEβ7インテグリン発現、CD3ε鎖の転写、TCR遺伝子の再構成などが引き起こされることを明らかにした。
