自己反応性B1細胞の異常増殖分化に基ずく疾患とその修復

研究概要	全身性エリテマトーデス(SLE)は幾つかの感受性遺伝子の関与する多遺伝子疾患で、その特徴的病態の一つに高lgG血症を伴うB細胞異常活性化が挙げられる。我々は、SLE自然発症モデル系である(NZB × NZW) F1 マウスをNZWに退交配したマウスを用いたマイクロサテライトDNA多型を利用した量的形質遺伝子(auantitative trait loci : QTL) 解析法により、高lgG血症の感受性遺伝子の遺伝子座位を決定した。その結果、これがNZBマウスの第1染色体テロメアのFcgr2b遺伝子に極めて密接に連鎖して存在することが明らかとなった。本年度は、Fcgr2b遺伝子が高lgG血症の血症の感受性遺伝子として機能しているか否かを知るために、NZBとNZW間でのFcgr2b遺伝子多型の有無を解析し、以下の点を明らかにした。 (1)NZBマウスのFcgr2b遺伝子プロモーター領域には正常マウスならびにNZWマウスには見られない13塩基ならびに3塩基の欠損部位が認められ、前者にはS box ならびにAP-4結合consensus sequence が存在していた。(2)これらの欠損によりNZBおよび(NZB×NZW)F1マウスでは、Fcgr2b遺伝子の遺伝子産物の一つであるFcγRIIB1分子の活性化胚中心B細胞上での発現が低下していた。(3)(NZB×NZW)F1×NZW退交配マウスの血中lgG量を、Fcgr2b遺伝子プロモーター領域がNZB型を持つ群と持たない群に分けて比較したところ、前者で有意に高値を示すことが明らかとなった。(4)B細胞上に発現するFcγRIIB1分子は、B細胞の活性化を抑制する働きを有している。従って、活性化B細胞上でのFcγRIIB1分子の発現低下を伴うNZB型のプロモーター領域多型は、NZBならびに(NZB×NZW)F1マウスに認められる高lgG血症の感受性遺伝子の一つとして機能していることが強く示唆された。
