ポリコーム群遺伝子Mel-18とBmi-1の前後軸形成における機能

研究概要

ほ乳類ポリコーム群のひとつであるMel-18とBmi1を欠損したマウスを用いた解析から、Mel-18とBmi1は初期リンパ球や胎児期の細胞増殖に必須であることとそれが細胞死の抑制を介する機序であることとHox遺伝子群の発現ドメインの維持に必須であることを明らかにした。このようなポリコーム群タンパクは、セントロメア領域に隣接して巨大なタンパク複合体を形成して機能することを示唆し、この複合体は細胞周期依存的にクロマチンから解離することを明らかにした。一方、Mel-18結合タンパクのスクリーニングを行なうことにより、ポリコーム群と細胞死制御のメカニズムの間を媒介する候補分子として、Ring1B、YAF2、Mph2、p53結合タンパクなどを同定した。Ring1Bの過剰発現または逆に発現量の低下は、Hox遺伝子群の発現ドメインの変化をもたらすことを明らかにした。Mel-18、Ring1B、YAF2とMph2は、タンパク複合体として核内に存在していることを強く示唆した。
