セレクチン、CD44の共通のリガンド・バーシカンの炎症細胞、癌細胞浸潤への関与

研究概要	われわれはこれまでに、白血球接着分子L-セレクチンとCD44に着目し、その機能解析およびリガンドの探索を行ってきた。その結果、CD44の新規リガンドとしてセルグリシンを同定するとともに、腎においてL-セレクチンのリガンドが遠位尿細管上皮細胞に局在することを報告した。さらに、ラット尿管結紮モデルにおいて腎間質への白血球浸潤が抗L-セレクチン抗体により強く抑制されることから、腎組織内への白血球浸潤にはL-セレクチンとそのリガンドの相互作用が重要な役割を果たすと考えられた。次に、腎尿細管由来細胞株ACHNの培養上清よりL-セレクチン・リガンドの精製を行った結果、既知のリガンドとは異なるプロテオグリカン・バーシカンを新たなL-セレクチンリガンドとして同定した。また精製バーシカンは、L-セレクチン以外にも炎症細胞浸潤関連分子であるCD44および種々のケモカインとも特異的に結合した。これまでL-セレクチンのリガンドとしてシアリルルイスX様糖鎖を持つムチン様糖タンパク質が報告されてきた。一方、バーシカンはコンドロイチン硫酸プロテオグリカンであり、既知のリガンドとは糖鎖構造、性状、分布が大きく異なる。このことから、バーシカンは、既知のL-セレクチン・リガンドとは異なる機能を有する可能性が考えられる。
