SH3ドメインをもつアダプター蛋白質の活性化と高次構造変化のメカニズム

研究概要	食細胞NADPHオキシダーゼの酵素本体は、細胞膜のシトクロムb_<558>(b_<558>)である。本酵素の活性化には、特異的アダプター蛋白質(p47^<phox>,p67^<phox>とp40^<phox>:それぞれSH3ドメインをもつ)と低分子量G蛋白質Racが刺激依存性に細胞質から細胞膜に移行してb_<58>と相互作用する必要がある。平成11年度は以下のような成果を得た。 (1)b_<558>と細胞質因子の相互作用は、p47^<phox>SH3ドメインとp22^<phox>(b_<558>の小サブユニット)の細胞質領域との結合に依存するが、p47^<phox>C末領域の3つのセリン残基のリン酸化が、SH3ドメインのunmasking、p22^<phox>との結合、及びオキシダーゼ活性化能誘導に必要であること、更にアラキドン酸がリン酸化と協同的に働いてp47^<phox>の構造変化を引起しオキシダーゼを活性化することを示した。 (2)Racの活性化を検出する新しい測定法を開発し、好中球刺激時のRacの上流で作用するシグナル分子を同定した。Racは、p67^<phox>のN末領域に存在するTPRドメインを介して作用し、オキシダーゼを活性化すること明らかにした。 (3)p40^<phox>は休止時細胞でp67^<phox>と会合しているが、これは新規なドメイン間(それぞれPCモチーフとPB1ドメイン)の新しいタイプの蛋白質間相互作用によるものであることを示した。PB1ドメインは酵母のシグナル伝達蛋白質Bem1pにも存在し、その役割を明らかにすると共に、その3次構造決定に成功した。 (4)p47^<phox>のN末領域に新規ドメイン(PB2ドメイン)を見い出し、その3次構造決定すると共に、PB2ドメインがPIP結合ドメインであり、p47^<phox>の膜移行に必須であることを明らかにした。
