低分子CXCR4アンタゴニストT22を基盤構造とした抗エイズ治療薬の開発研究

研究概要

本研究は、T-細胞指向性HIVの第二受容体であるケモカインレセプターCXCR4のペプチド性拮抗剤T22を基盤分子としたエイズ治療薬の開発を目的としている。本年度は、低分子CXCR4拮抗剤の開発に関連して、我々はT22の活性コンフォメーションを保持したまま分子サイズを低減化した14残基ペプチドT134及びT140の作用機序の厳密な評価を行うとともに、T134がDe Clercqらの見いだしたCXCR4拮抗剤AMD3100と交差耐性を示さないことを明らかにした。さらに、T134及びT140を新たなリード化合物とする分子サイズの低減化及び非ペプチド化に重点をおいた合成研究を実施した。特に非ペプチド化研究においては、我々の研究室で詳細な検討を行ってきた遷移金属触媒Pd(0)によるビニルアジリン系化合物の異性化反応、有機銅試薬RCuLnによる立体特異的anti-SN2'反応に加えて、Grubbsらにより開発されたルテニウム(Ru)触媒を用いるオレフィンメタセシス閉環反応等の有機金属触媒の特性を有効に活用した合成化学的アプローチを実施した。これらの方法を応用して、T140のジスルフィド結合を(E)-Alkene結合に変換し、type II β-turn構造中の(i+1)番目及び(i+2)番目にDLysψ[(E)CH=CH]Alaを導入したTM(E)140は高い抗HIV活性(EC50=11.5 nM)と優れた選択係数(SI=22763)を示した。現在、TM(E)140を基盤分子として設定し、さらなる非ペプチド化、ダウンサイジングを実行中である。
