機能性核酸のDNA複合体形成によるシンクロナイゼーション活性化

研究概要	背景:合成オリゴヌクレオチドは生体内においてmRNAあるいは2本鎖DNAと複合体を形成することによって、特異的に遺伝子発現阻害を行うことが可能であり、アンチセンス、アンチジーン法として新時代の医薬品への発展が期待されている。高い阻害効率を実現するため、これらの複合体の安定化が必要であり、特に、機能性核酸を利用した核酸間を共有結合で固定するクロスリンク反応の適用が検討されている。我々は、効率的かつ特異的な反応が、目的の標的部位でのみ活性化される分子の開発を目指し、標的核酸との複合体形成→活性化→クロスリンク反応と一連の反応がシンクロナイズして誘起される、新規機能性核酸誘導体の開発を目指している。平成11年度の成果:反応基としてフェニルするフォキシドを有する2-aminopurine誘導体を用いるとき、約100%近い収率で、シチジン選択的および部位特異的に反応する優れた反応であることを報告した。今回、スルフィド芳香環上の置換基が電子吸引性基は反応効率を減少させ、電子供与性基が反応収率を大きく向上させることを見出した。そこで、よりより電子供与性基としてカルボキシルレートの置換体を検討したところ、活性化体としてのビニル体に近い反応収率が得られた。グルタチオンおよびアスコルビン酸など生化学的なラジカルスキャベンジャーの添加によりスルフィド体のクロスリンク反応が完全に阻害され、スルフォキシド体の反応は影響を受けなかったことから、このシンクロナイゼーション活性化には、電子移動が鍵反応として含まれることが示唆された。現在、中性条件下でも機能する効率的なクロスリンク剤についても検討中である。
