研究概要	キネシン分子モーターの"歩く"仕組み:キネシン分子が微小管上を"歩く"仕組みについては、単頭結合と双頭結合とを繰り返すというhand-over-handモデルが提唱されている。ところが、直接的な証拠を提出すべき電子顕微鏡観察の結果が互いに矛盾しており、新たな証拠が求められていた。そこで我々は、キネシン分子と微小管との1分子結合力(結合破断力)を、1)ATP非存在下、2)AMP-PNP(ATPのアナログ)とADPの共存下、そして、3)AMP-PNP存在下という、hand-over-handモデルにおける鍵となる3つのヌクレオチド状態で顕微計測した。その結果、1)と2)での平均破断力が7pN、3)での平均破断力が15pNと求められた。しかも、各条件下でのキネシン分子の弾性率が、それぞれ約0.4pN/nmと約0.8pN/nmと見積もられた。これらのことから、前者では単頭結合、後者では双頭結合であると結論でき、hand-over-handモデルが強く支持された。このことは、キネシン分子モーターの歩く仕組みの中に"分子内双頭間分子協調性"が存在することを強く示唆する。つぎに、1分子のキネシンが発生する力、歩く速度、一度に歩く距離、の3つの特性に対する温度(15-35℃)の影響を検討した。その結果、意外なことに発生力は温度に依存せず約7pN、歩く速度と歩く距離は温度の上昇とともに増大した。これらの結果は、歩く仕組みの解明に示唆を与える。心筋収縮系に見られるSPOC現象:昨年までの研究に加えて、ラットのグリセリン処理心筋収縮系について、ADP-SPOC溶液中でのSPOC周期などを顕微計測した。その結果、SPOC周期は動物種によらず各心拍周期の約24倍であることが分かった。このことは、心筋収縮系自体、自発的な振動系であり、心拍リズムを担う土台として適応していることを示している。
