昆虫行動の中枢性補償と微小脳システムの可塑性

研究概要	前年度までの調査は、片側尾葉切除を受けたコオロギが空気流刺激に対する逃避方向を補償するためには、自由に動くことのできる環境が必要であることを明らかとした。このことから、コオロギの微小脳システムは自身の動きから期待される空気流感覚系への自己刺激と、実際の引き起こされる自己刺激との不整合性を手がかりに巨大介在神経(GI)群の機能補正を行っているという仮説をたて、それを検証する方向で研究を進めた。自己刺激量を予測するために必要な、自発歩行時の運動出力のコピーシグナル、すなわちEfference copyに相当する情報をGI群に伝えているであろう下行性の介在神経の同定を試みた。まず、腹部縦連合において下行性の神経群を電気刺激し、上行性の神経に反応を引き起こす割合を調査したところ、約30%の標本でGIの反応が確認された。下行性介在神経のインパルス伝導速度を推定したところ、平均1.4m/sとなったことから、それらは直径1-2μmの比較的細い軸索を持つ介在神経であることが判明した。続いて、自発歩行中に腹部縦連合から下行性の神経活動を記録し、それらの発火パターンについての解析を行った結果、自発歩行中にリズミカルなバーストを示す下行性介在神経が存在することが判明した。細胞外記録によるインパルスの振幅は小さいことから、それらは細い軸索を持つ神経であると考えられ、電気刺激実験による結果と一致した。また、それら下行性介在神経のバースト頻度は歩行速度すなわち自己刺激量をコーディングしていること、また左右の下行性介在神経の同時記録から、それらのバースティングパターンはコオロギの歩行の方向をもコーディングしていることが判明した。これらのことから、それらの下行性介在神経は自発歩行時の運動出力のEfference copyを腹部最終神経節へと伝え、神経系の補償的機能回復に関与している可能性が強く示唆された。
