研究概要

本研究は、シグナル伝達における蛋白質複合体の詳細な構造・機能解析を目指して、以下の2点を目指している。(1)抗ニワトリリゾチーム抗体HyHEL10Fv-エリスロポエチン受容体キメラ蛋白質を受容体のリガンド認識機構研究のモデルケースと捉え、HyHEL10Fv-受容体キメラ蛋白質と抗原リゾチームとの複合体のX線結晶解析を行う。(2)異種抗原認識能を有するHyHEL10Fv変異体を用いて細胞増殖能の変化を調べ、さらに変異HyHEL10Fv-受容体キメラ蛋白質とリガンド複合体の構造解析を行い、分子認識能と細胞表面からの情報伝達の機構に迫る。今年度の実績は以下の通りである。(1)まず、ヒト並びにマウス由来のエリスロポエチンについて、その細胞外ドメイン遺伝子をクローニングし、大腸菌を用いた高発現系を確立した。抗体分子に準じた、不溶性顆粒からの高効率な巻き戻し系により、100%の効率での巻き戻し系を構築した。HyHEL10のVHまたはVLとマウス並びにヒト由来エリスロポエチン受容体の細胞外ドメインを置換したキメラ蛋白質について、その大腸菌を用いた高発現系並びに巻き戻し系を構築することで、X線結晶解析をはじめとする各種物理化学的解析に充分対応できる試料を供給する系を確立した。(2)既に確立してあるFvとニワトリリソチームとの複合体の結晶化条件をもとに、次年度における抗体-受容体キメラ蛋白質の構造機能解析において用いる変異抗体について、抗原との複合体の結晶を作製し、高エネルギー加速器研究機構の放射光装置で精密なデータ収集を行い、相互作用について詳細な考察を行った。
