立体構造に基づく蛋白質合成マシーナリーの動作機構の解明

研究概要	本研究の目的は、遺伝情報の変換過程(複製、転写、翻訳)に携わる酵素およびRNAの高次構造を決定し、生物マシーナリーとしての動作機構を解明することにある。本年は、まず高度好熱菌由来のグルタミルtRNA合成酵素(GluRs)とtRNAGluの複合体の結晶構造を2.4Å解能で決定し、tRNAのアンチコドンの認識機構を解明した(Nature Structural Biol.,2001)。本構造では、tRNAGluのアンチコドン3字目のC36がArg358により水素結合で認識されていたが、このArg残基をGlnに置換することで、変異体GluRSはtRNAGln型のアンチコドン(34YUG36)を認識する非識別性GluRSに変換されることがわかり、わずか1残基のアミノ酸置換でtRNAの認識が変換されるという分子進化の過程を実証することに成功した。また、高度好熱菌由来アルギニルtRNA合成酵素(ArgRs)の2.3Å分解能の結晶構造を決定し、系統的な変異体解析の結果、tRNAArgのA20の特異的な認識機構を解明した(Proc. Natl. Acad. Sci. USA,2001)。また超好熱古細菌Pyrococcus horikoshiiのリジルtRNA合成酵素(LysRS)(真核生物・真性細菌ではクラスIIの酵素が存在するが、古細菌由来の本酵素はクラスIである)の結晶構造をMAD法により2.6Å分解能で決定し、構造の全く異なる2種類のLysRSが同じアミノ酸、tRNAを認識するメカニズム(機能的収歛)を明らかにした(Nature Structural Biol.,2002)。さらに、古細菌由来のtRNAのG15を、塩基置換反応によりqueuosine置換体に修飾するarcTGTと基質塩基との複合体の結晶構造を2.2&ェ解能で決定し、塩基置換反応のメカニズムを解明した(J. Mol. Biol.,2002)。また、リボソームRNAのメチル化を触媒する2'-O-MTaseの結晶構造を2.4Å分解能で決定し、本酵素ファミリーの触媒部位には明瞭な結び目(knot)構造があることを発見し、これが酵素活性部位の構築に重要であることを示唆した(論文投稿中)。さらにDNAの複製の際にRNA primerを合成するDNAプライマーゼの結晶構造を1.8Å分解能で決定した(論文準備中)。また、セントロメアにおけるクロマチンの構築に必須なCENP-BとDNAの複合体の結晶構造を2.5Å分解能で決定した(EMBO J.,2001)。さらに、MRSAの特効薬であるmupirocinとその標的タンパク質であるイソロイシルtRNA合成酵素の複合体の結晶構造を2.5Å分解能で決定し、抗生物質の作用機序を明らかにした(J. Biol. Chem.,2001)。
