研究概要

PheB酵素の構造決定と基質特異性の起源の解析
Extradiol型のdioxygenaseであるPheB酵素の立体構造を、基質非結合型、基質結合型の両者に関して決定した。結晶の質が良くないため、分解能は2.8Åと十分とは言えないがextradiol型dioxygenaseの基質特異性には、サブユニットの会合の仕方やC末端部位のアミノ酸(Phe299)が関与していることを強く示唆する結果が得られた(投稿準備中)。
BphA4(ferredoxin reductase)の結晶構造の決定
PCB分解系中の初発酸化酵素であるBphAの電子伝達系に存在するBphA4(ferredoxin reductase)の立体構造を2.1Å分解能で決定した。また、BphA4-NADH複合体に関しても2.6Å分解能で、その立体構造を決定した。この結果、BphA4がGlutathione reductase(GR)に構造が類似していること、NADHの結合様式もGRのそれに類似していることが明らかになった。これらの立体構造から、BphA3(ferredoxin)の結合位置やBphA4内の電子伝達の経路に関して新たな知見が得られた(投稿準備中)。
BphA1A2(biphenyl dioxygenase)の構造解析
BphA1A2は、PCB分解系中の初発酸化酵素であるBphAの活性部位を構成するコンポーネントで、α_3β_3のサブユニット構成を持っている(分子量約20万)。本年度は、Phodococcus sp strain RHA1由来のBphAlA2の結晶化条件の最適化を行い、約3Åの回折点を与える結晶を得た(投稿準備中)。この結晶の空間群はP4_12_12で、格子定数は、a=b=139.9Å c=240.2Åであった。この結晶を用いて放射光実験施設(PF)において回折データの収集を行った。また、水銀の重原子同型置換体の作成にも成功している。このため、平成12年度には立体構造を決定できると考えている。
