研究概要	本特定研究の課題における我々の研究課題は、複数の反応中心をもつ可溶性フラビン酵素の構造と電子伝達について系統的な理解を得ることである。現在課題の中心であるキサンチン脱水素酵素は、尿酸代謝や活性酸素生成酵素として重要な酵素である。又、本酵素は、特定のSH基の酸化または、蛋白分解酵素による限定分解で基質特異性の変化を来す、つまり電子受容体をNADHとする脱水素酵素型から電子受容体を酸素とする酸化酵素型に変換することが知られている。昨年度は脱水素酵素型酵素が2.1Åの分解能で解けたが、それを基にして限定分解によって得た酸化酵素型の構造が2.5Åの分解能でとけた。フラビン周囲の構造を詳細に検討した結果、脱水素酵素型と酸化酵素型酵素の構造上の差異が明らかになった。脱水素酵素型で存在したフラビンドメインとモリブドプテリンドメインの間を結合する長いリンカーが酸化酵素型では切断されるため、それに連動したフラビン平面に位置するループが限局しながらも、大きくフラビン上方に移動しNADの結合を妨げていた。と同時に、そのループの移動でフラビン周囲のイオン環境が大きく変わり、フラビンセミキノンを取りにくい環境、つまり酸素と反応しやすい状態になって酸化酵素型に変換したと考えられた。これらの構造的変化は今までの生化学的な種々な実験結果を支持する。また従来より広く使用されている抗痛風剤であるアロプリノール、新薬、TEIとの共結晶が得られたので、更に高分解能の解析を試みている段階である。
