研究概要	本申請は生物マシーナリーの結晶が抱える不安定性、回折強度の弱さを克服して精度の良いデータ収集環境をフォトンファクトリー(PF)のユーザー全体に提供し、高精度の構造解析を可能にすることを目的としている。具体的には以下の3点が検討する計画である。 1.最適条件でのデータ収集放射光実験の直前にオフラインで予備実験を行える環境(X線回折装置)を実現して、解析の高効率化を実現する。 2.回折データ収集の高速、高精度化生物マシーナリーの解析に用いられるMAD法およびファインスライス法を可能にするため、データ収集の律速であるIP読み取り時間の短縮を実現する。 3.多波長異常分散法に向けた試料の調製及び結晶化現在解析中の蛋白質(精巣上体分泌蛋白質)の結晶化を行う。今回導入するX線回折装置を用いて、結晶の評価、低温実験の条件の検討を行う。本年度1と3の項目を達成するために、X線回折装置の購入を行う予定であったが、CCDを用いた2次元検出器の性能がタンパク質結晶構造解析に十分使用可能であり、回折データ収集の高速化が図れるとの考えから、本予算でCCDを用いた2次元検出器の購入を行うこととし、仕様の決定等を行った。平成12年はシンクロトロン放射光を用い検出器の評価実験を行う。
