研究概要

海馬、嗅球、新皮質のいわゆる広義の大脳皮質を構成するニューロン群、主に、GABAニューロンについて分布密度等の定量解析、GABAニューロンサブポピュレーションの化学的、形態学的性質の解析を進めた。平成11-12年度はGABAニューロン群の特徴あるサブポピュレーションであるparvalbumin(PV)含有GABAニューロンの形態学的性質、特に、PVニューロン相互の関係について海馬の及び新皮質で研究を進めた。PVニューロンは錐体細胞の主として細胞体およびaxon initial segmentに抑制性シナプスを作り、活動電位の発生を直接調節する役割をもつと考えられている。PVニューロンはまた、お互いの細胞体の上にも密にGABAシナプスを形成することで介在ニューロンの相互抑制回路を形成し、かつ樹状突起間にgap junctionを形成する。これらの形態学的特徴は、多数の神経細胞がリズミカルでかつ同期した活動を行う同期的振動、特に40Hz付近のガンマリズム発生の機序を説明する生理的所見及び理論モデルに合致している。興味深いことに海馬PVニューロンの遠位樹状突起は予想外に側方に広がり、かつお互いに非常に密に接触し、海馬全体に広がる樹状突起ネットワークとも言うべき構造を作っていた。電子顕微鏡と共焦点レーザー顕微鏡を用いた検討で、この樹状突起ネットワークがgap junctionを介してつながっていることを明らかにした。これは、細胞体が離れて存在するPVニューロンが、位相のそろった入力を受ける可能性を示唆する。
更に、新皮質の各領域においても、PVニューロンは相互に化学的シナプスを作り、その樹状突起は海馬同様に多数の個所でお互いに接触し、その接触部位にgap junctionの存在が確認できた。ただし樹状突起の広がりが、海馬では常に側方へと大きく展開しているのに対して、新皮質では、領野、層および種に特異的な方向性を示し、機能的性質との対応が予想される差異が認められた。海馬、新皮質のPVニューロンで明らかにした、化学シナプスと電気シナプスを介する二重のネットワーク構造は、in vivoで観察される広範囲での同期的振動の発生機序を探るうえでの、形態学的基礎となりうる可能性を有している。
