ポリ乳酸をベースとする生分解性ポリエステルの構造と物理的特性に関する研究

研究概要

13年度は、主に以下の4つの項目に関する研究を行った。
1.ポリ(L-乳酸)の酵素的加水分解挙動の高次構造依存性 ポリ(L-乳酸)(PLLA)フィルムのproteinase Kによる酵素的加水分解速度は、結晶化度(x_c)<30%では、結晶化度の増加とともに著しく低下するが、x_c≧30%においては、低下が緩やかになることがわかった.これは、x_c<30%では、分解速度の高い球晶外のfreeな非晶領域が主に加水分解されるが、x_c≧30%では、分解速度の低い球晶内の結晶領域間の非晶領域が主に分解されることによる。
2.ポリ(L-乳酸)の高温加水分解挙動および高次構造依存性 PLLAのリン酸緩衝液(97℃)中の加水分解挙動においては、体温における結果と比較して高次構造の影響は小さいが、結晶化度が高いほど分解速度が高いことを明らかにした。
3.ポリ乳酸の酵素的加水分解に与える分子構造および光学異性体間のポリマーブレンドの影響 PLLAのproteinase K存在下での加水分解速度は、分子量の低下とともに増大すること、および分解は、endo的およびexo的に進行することが明らかとなった。L-乳酸含率が30%以上、L-乳酸連続単位数が3以上、D-乳酸連続単位数が10未満であれば、proteinase Kによる分解は有意に加速された。また、4より多いD-乳酸連続単位の存在が、L-乳酸連続単位の分解を阻害することが、明らかとなった。
4.酵素を用いた多孔化 proteinase Kを用いた場合、ブレンド比[PLLA/ポリ(ε-カプロラクトン)(PCL)]が50/50または30/70であれば、PLLA/PCLブレンドから100%PLLAが分解・除去された多孔化PCL材料が5日後に得られたが、lipaseを用いた場合には、12日間以上分解を行ってもPCL相の分解・除去は不十分であった。
