研究概要

種々のリパーゼを用いて天然に存在するひまし油から得られる12-hydroxystearic acid(12-HAS)の酵素重合を検討した。その結果、Candida antarctica由来のlipase CAが最も高い重合活性を示した。この反応をベンゼン中80℃で一週間続けることによって12-HASの環状二量体(13,26-dihexyl-1,14-dioxacyclohexacosane-2,15-dione,12-HAS)が優先的に生成されること見出した。lipase CAによる12-HASの重合では、直鎖状のoligomerが生成し、その後、back-biting反応が起こり、大環状の12-HASDが形成される反応機構が考えられる。
diethyl zincを触媒に用いて12-HSADのバルク重合を行った結果、12-HSADは120℃付近に床温度を有する開環重合性を示すことを認めた。そこで、12-HSADをまず重合させた後に、L-ラクチドを添加してブロック共重合を行い、12-HSAD単位をPLLAに効率的に導入することに成功した。
PLLAとPDLAはステレオコンプレックスを形成することが知られている。このステレオコンプレックス形成を共重合体間の会合制御に利用して新しい水系ゾル-ゲル転移システムを構築した。すなわち、PEG-PLLA-PEG、PEG-PDLA-PEGトリブロック共重合体を水中に分散させて、微粒子分散液を調製し、低温で両者を混合したところ、混合後徐々に粘度が増大しゲル化が生じた。このゲルを加熱するとゾル化し、逆に温度を下げると再びゲル化を生じたことより、ゲル化はPLLAとPDLAのラセン形成によって誘導されたラセン状PEG鎖の会合によって進行していると考えられる。これは、新しいゲル化機構である。
Poly(butylene succinate)(PBS)に芳香族単位としてテレフタル酸単位を共重合したpoly(butylene succinate-co-terephthalate)(PBST)およびpoly(butylene adipate-co-terephthalate)(PBAT, Ecoflex^<【○!R】>)の酵素分解性について詳細に調べ、分解生成物の分析から分解機構を検討した。
