研究概要	環境ならびに各種ストレスに応答する細胞の調製、遺伝子の探索を目的として以下の結果を得た。(1)NF-κB結合領域の下流にルシフェラーゼ構造遺伝子を連結した遺伝子を導入したヒト肺癌腫細胞を構築し、IL-1あるいはTNFの刺激によるNF-κBの発現を発光の程度により検定することを可能にした。(2)大腸菌の染色体分配機構解析のために、抗FtsZ抗体を用いた分裂環の免疫蛍光染色とDAPIによる核染色を同時に行ない、染色体分配と細胞分裂の同時観察を可能にした。(3)低周波微小電圧刺激により活性化されるプロモーターについて検討し、heat shock proteinのひとつであるhsp70が該当することを見出した。(4)hsp70プロモーターの下流にルシフェラーゼ遺伝子を連結した動物細胞発現ベクターを構築し3T3-L1細胞に導入することによりhsp70の発現を発光により定量化することに成功した。さらにこの細胞を使用して熱刺激と電圧刺激ではhsp70遺伝子の発現経過に差異があることを示した。(5)有機溶媒に曝露したり酸化状態にすると発現が誘導される大腸菌遺伝子であるsox-marレギュロンよりsoxR遺伝子のプロモーター領域を取得し、下流にgreen fluorescence protein(gfp)遺伝子を連結したプラスミドを調製した。このプラスミドを導入した大腸菌は、スーパーオキシド生成試薬methylviologenの存在下で蛍光を発し遺伝子発現を容易に検出することを可能にした。(6)S. violaceaDSS12の加圧応答プロモーター領域にσ^<54>因子の結合部位があることからσ^<54>因子の遺伝子クローニングを行ない、加圧下発現遺伝子のひとつであるグルタミン合成酵素遺伝子のプロモーター領域との結合を検討した。(7)リポソーム中でアクチンを成長させた際、束化能をもつα-アクチニンの存在によりアクチン単独の場合とは異なり、球状部分の両側に管状部分を持つ二極性リポソームが形成されることを観察し、生体内ストレスファイバーのモデルとなることを示した。
