研究概要	低周波微小電圧刺激に応答するプロモーターの下流に神経細胞特異的転写因子NeuD2遺伝子と緑色蛍光蛋白質(GFP)遺伝子を連結したプラスミドを作成し、神経芽腫瘍細胞株(N1E-115)を形質導入した。この細胞は、電位印加により分化誘導され、緑色蛍光を呈するとともにその形態を神経細胞様に変化させた。(相澤) 大腸菌の多剤耐性に関与するmarRABオペロンの発現調節領域と、marRABオペロンの転写抑制蛋白質をコードするmarRのクローニングとともに、marA遺伝子の5'末端領域にGFP遺伝子を融合した。ミスセンス変異を有するmarRを用いた融合遺伝子を有するプラスミドで形質転換した大腸菌は、芳香族化合物の濃度に応じて蛍光を呈した。(青野) テキンがFenton反応を誘導してHydroxyl radicalを発生させることにより活性型破骨細胞のアポトーシスを特異的に誘導することを見いだした。また細菌の染色体分配を阻害し、細胞の球状化を誘導して無核細胞の放出を引き起こす、S-Benzylisothiourea骨格をもつ化合物を発見した。(和地) プラスミド上のlacプロモーター制御下のGFP遺伝子が30MPaの加圧条件下にて特異的に発現誘導され発光する高圧応答大腸菌の構築に成功した。また、好圧菌の加圧応答する転写においてNtrBが圧力センサーとして働く可能性を示し、より高圧条件(50~70MPa)を認識する高圧応答細胞の構築の可能性を示した。(加藤) アクチン結合タンパク質であるファシン、α-アクチニン、フィラミンをG-アクチンと共にリポソームに封入すると、より複雑な構造が形成されリポソームを特異的な形に変化させた。またリポソームにラテックスビーズを平均数個取り込ませる方法を確立し、ダブルビームのレーザーツイーザーシステムにより機械的張力を加え、その形態応答を可視化・制御した。(宝谷)
