金属錯体の合理的設計による新しい酸素分子活性化反応

研究概要

酸素分子の活性化を行う金属酵素としてシトクロムP450のモデル構築による反応活性等について研究を進めた。前者の酵素については、ヘムの両面の嵩高いビナフタレン基を架橋させ酸素配位部位近傍に水酸基を有する「双冠ボルフィリン」を用い、ビナフタレン基により形成される一方のホケット内にアルカンチオレート基を共有結合により固定した欽(III)錯体が空気中において安定に生成することを明らかとした。さらに、還元体に対する酸素錯体の形成に成功し、モデル錯体としては世界で初めて0℃程度まで安定なチオレート配位酸素錯体形成を行い、その構造を詳細に共鳴ラマンスヘクトル等により解明した。この錯体においては配位酸素は一方のホケット内にある強い電子供与性基アルカンチオレートにより蓄積した末端酸素原子上の負電荷を近傍に存在する水酸基からの水素結合により中和することにより酸素錯体が安定化されているものと解釈された。またこの酸素錯体は、一酸化炭素(CO)と可逆的に配位子交換を行い、特徴的なスペクトルを与える。しかし、配位した一酸化炭素の振勤スペクトルν(Fe-C)は他のヘム類縁体と比べて異様に低いことが明らかとなり、COに対する親和性も大きく低下していることが見いだされた。そこでこれらのヘム含有酵素における酸素配位部位の構造は、酸素運搬機能を有する金属酵素ミオグロビン、ヘモグロピンとも類似していることから、この双冠ボルフィリンに対するCO配位の特性の一般性を解明した結果、配位したCO近傍に存在する水酸基酸素原子上の非結合性電子対により生じる電子的な影響によりCO配位体が大きく不安定化されている事を世界で初めての実験的に立証した。
