研究概要	活性中心が鉄ポルフィリン錯体(ヘム)によって構成されるタンパク質・酵素をヘムタンパク質・ヘム酵素と呼ぶ。ヘムは93番目のアミノ酸であるヒスチジンに配位することで、蛋白内部に取り込まれている。本研究では、活性中心を構成するヘムを他の金属錯体に置換し、ヘムタンパク質・ヘム酵素とは異なる機能の発現を目指した研究を行っている。平成12年度は、ヘムの位置に配位しうる様々な金属錯体の合成を行った。その最初のシリーズは、鉄、マンガン、コバルトを導入したシッフ塩基錯体である。本研究に類似した研究がこれまでに2報報告されているが、文献に従った実験を行い、蛋白内部に錯体が挿入されているか否かを検討したところ、93位のヒスチジンに配位しておらず、直ちに蛋白外に放出されると言う結果であった。これに対し、我々の合成した金属シッフ塩基錯体は蛋白内部で93位のヒスチジンと配位結合により安定に取り込まれていることが、マススペクトルの測定から明らかになった。従って、安定に金属錯体を蛋白内部に導入した人工金属タンパク質合成の初めての例と言うことになる。得られた人工金属タンパク質に、価額返還を触媒する能力があるのか(すなわち、酵素と呼べるのか)を検討するために、酸化反応を行った。その結果、酵素活性は未だ低いものではあるが、不斉酸化をスルフィドに対し達成することが出来た。現在、他の反応に関する触媒活性を検討しているところである。一方、マンガンを導入したポルフィリン錯体の導入を行い、過酸との反応を検討した。鉄に比較して反応は遅く、活性も低いことが分かったが、反応性が緩衝水溶液のpHに非常に敏感であり、pH5以下では非常に高い反応活性が現れることが明かとなり、現在反応条件下の最適化を検討中である。
