体内時計の光位相同調とメラトニン出力系の調節メカニズム

研究概要	交付申請書に記載の研究目的に従い本領域では、ニワトリやラットの松果体における体内時計の光入力系・メラトニン出力系についての研究を遂行し下記の成果を得た。 (1)ニワトリ松果体において光受容体と連関する2種類のG蛋白質(GtおよびG11)の全長cDNAおよび、時計遺伝子の候補分子群の部分cDNAの単離に成功した。(2)松果体において光刺激に伴いmRNA量が変動する複数の遺伝子を単離した。(3)松果体・網膜においてオプシン型光受容体はG11を介して光情報を概日時計に伝達し、位相同調を担っていることを明らかにした。(4)光による転写活性化の分子解析を進めるため、光依存的な転写誘導に関わるエンハンサー領域を同定した。(5)ラット松果体におけるグルタミン酸を介したメラトニン合成抑制経路の解析を進めるためグルタミン酸受容体を網羅的に検索した。その結果、代謝型受容体mGluR5およびイオン型受容体・AMPA型GluR1が発現していることを突き止めた。(6)グルタミン酸がラット松果体においてNATのみならずHIOMTをも阻害することを見いだした。(7)ラット松果体細胞におけるセロトニンの合成・分泌機構を調べた結果、Vesicular monoamine transporter 1が関与するという証拠を得た。(8)グルタミン酸信号の終止系として機能する形質膜型のNa^+-dependent glutamate transporterのcDNAをラット松果体より単離した結果、新規のイソフォームであった。以上のように本研究では、脊椎動物の概日時計の入出力系について数多くの知見を得ることができた。今後、発振分子と直接相互作用する入力分子の同定を進めると共に、peripheral glutamatergic systemの全貌解明、未知なるセロトニンの動態解明が大いに期待できる。
