研究概要

精子形成の分子機構を解明するために、半数体精子細胞に焦点をしぼり、特異的に発現する遺伝子群cDNAの包括的なクローニングを行った。それらにコードされる蛋白質の局在と生化学的な解析により、半数体細胞では精子という特殊な最終形態への分化に必要な特殊な分子が積極的に発現していることが明らかとなった。さらにこれらの遺伝子のゲノム構造を解析したところ、イントロンレス遺伝子が多く、その発現制御には特異的な転写因子による制御だけではなく、生殖細胞特異的なクロマチン構造の変化が重要であることが示された。遺伝子発現におけるエピジェネティックな制御は、個体の発生過程で重要となるが、その獲得は生殖細胞の分化・成熟過程に行われる。これまで、エピジェネティクスの解析は、インプリンティング遺伝子を中心に行われてきているが、今回クローニングされた半数体精子細胞特異的遺伝子群は、生殖細胞と体細胞間でのクロマチン構造の違いと生殖細胞特異的な制御についての解析ツールを提供するものである。今後、ジェノミックインプリンティングの獲得機構も含め、生殖細胞におけるエピジェネティクス解析への可能性が期待される。
一方、精子形成の異常は、ヒトにおいては不妊症として捉えられるが、その原因の半数が男性側にあるとされるにもかかわらず、その本態は不明であるものがほとんどである。しかし、今回の成果からヒト男性不妊症の遺伝子レベルでの解析の可能性が示された。今後、内分泌攪乱物質などスクリーニングを始め、男性不妊症の遺伝子診断法の確立など研究成果の社会還元へ向けた努力が必要である。
