研究概要	ウイルス感染が起きた細胞内では、はじめにIFN遺伝子を含む種々の遺伝子の発現が誘導され、抗ウイルス作用などの生体防御機構が働く。我々はこの一連の活性化機構のうちIRF-3が最初に活性化される転写因子であり、細胞質のシグナルを直接核へ伝達する事を明らかにしてきた。IRF-3モノマーはその分子内に存在するNuclear Export Signal(NES)の機能により細胞質に局在し、しかも転写因子としての機能を持たない不活性型である。しかし、ウイルス感染の刺激にによってC端側に存在する特定のセリン残基が燐酸化されることにより2量体形成する。p300/CBPはIRF-3の2量体と選択的に複合体を形成し、DNA結合能を獲得するが、この時、p300/CBP自身がDNAと相互作用するという事を発見をした。本年度は複合体形成の再構成系を用い、p300/CBPの機能ドメインを解析した。その結果、IRF-3の2量体とp300/CBPの相互作用はp300/CBPのグルタミンに富む領域がインターフェースとして機能していることが明らかとなった。しかし複合体形成と同時に観察されるDNA結合能にはさらに別のドメインが必要であり、その一つはHATドメインであった。しかも、点変異による解析の結果、HAT活性自身が必須であること、また、IRF-3が複合体中でアセチル化されることが判明し、この新規な化学修飾の重要さが強く示唆された。第3のドメインはCH3と呼ばれる領域で、その中でも各種の転写因子と相互作用する事が報告されている、TAZ2ドメインが必須であった。この領域のみでDNAと相互作用しうる事から、IRF-3ホロ複合体の特異的DNA結合能にこの領域が関与していることが強く示唆された。以上のようにIRF-3は分子の燐酸化、アセチル化等の修飾並びに他の蛋白質との相互作用によって不活性型から活性型へ転換する事が明かとなった。
