バラ科植物における配偶体型自家不和合性の分子機構

研究概要

自家小和合性は、近親交配を回避し植物集団内における遺伝的多様性を保持するため、雌しべが自己と非自己の花粉を認識し非自己花粉でのみ受精する性質である。この性質は1遺伝子座の複対立遺伝子(S遺伝子)により支配され、雌しべと花粉でそれぞれ特異的に発現する蛋白質(S蛋白質)の分子間相互作用により雌しべ・花粉間の自他認識が行われると考えられている。近年、花粉特異的に発現しているF-box遺伝子がS遺伝子座に発見され、花粉側S蛋白質である可能性が強く示唆されている。しかし、F-box蛋白質がS-RNaseと相互作用する直接的な証拠はなく、F-box蛋白質が関与するユビキチン・プロテアソーム系が配偶体型自家不和合性の本質である証拠も未だ示されていない。そこで本研究では、バラ科ニホンナシを研究対象に、S-RNaseおよびF-box蛋白質の細胞内動態の解析から配偶体型自家不和合性の分子機構の解明を目指した。
まず、精製したS-RNaseとハンギングドロップ法により再現性よく花粉管伸長阻害が起こる実験系の確立に成功した。次に、この実験系を用いて花粉管伸長阻害時に特異的に発現している蛋白質およびS-RNaseの構造変化を解析した。その結果、花粉管伸長阻害時に特異的に発現している13種類のタンパク質が検出されたが、F-box蛋白質は検出されなかった。上記の実験において、花粉管内に取り込まれたS-RNaseは何らかの修飾を受け、S-RNase添加から12時間経過後には花粉管内に取り込まれたS-RNaseはすべて分解されていた。さらに、S-RNaseの代わりにin vitro花粉管伸長阻害実験系にRNase T2やRNase Aを添加しても、RNase自身の分解は観察されなかった。以上の結果は、S-RNaseを積極的に修飾・分解するシステムが花粉管内に存在していることを示唆している。
