研究概要

1.アブラナ科植物の自家不和合性の花粉因子の探索
アブラナ科植物の自家不和合性の花粉側の因子の候補分子として、これまでシステイン8個を含む分子量約7000のタンパク質を見いだしてきたが、本研究では、さらに、その候補分子に類似した遺伝子は全てのS系統の植物に存在し、それらのアミノ酸組成が大きく変化していること、また、ゲノム上SRKのきわめて近傍にあることを明らかにした。さらに、花粉や葯での発現すること、さらに、形質転換実験、および、柱頭上での花粉の吸水反応を指標とした生物検定系によって、これが花粉因子の本体であることを確定することが出来た。この遺伝子は便宜的にSP11と呼んでいる。現在、この花粉因子を大腸菌やバキュロバイラスによる発現、あるいは化学合成によって、大量に得ることを計画している。また、S遺伝子座の全体像の理解のため、この領域のゲノム解析をすすめている。
2.ペチュニアの自家不和合性にかかわるS遺伝子座の解析
ペチュニアは配偶体型自家不和合性を示し、柱頭のS遺伝子座はS-Rnaseである。本研究では、S-Rnaseを含むゲノム断片約40kbを得て、その塩基配列を決定した。また、その断片を使ったFISH法により、ペチュニアではS遺伝子は、第3染色体のセントロメアのごく近傍にあることを明らかにした。現在、この領域にあり、花粉で発現している遺伝子を探索している。
