真核細胞ゲノム上の複製ドメイン・レプリコンの複製機序の解明

研究概要

本研究では、ゲノムDNAの複製進行過程・核内配置・クロマチン構造のダイナミクスの視覚化により、核内ゲノムのダイナミクスに関する情報の蓄積を目指す。
1.出芽酵母の初期複製開始部位をDNAファイバー上で検出する方法を考案した。ブロモデオキシウリジン(BrdU)を取り込むことができる出芽酵母をalphaファクターでG1期後期に同調し、ヒドロキシウレア(HU)、BrdU存在下でS期にリリースする。HUはヌクレオチドの供給を阻害するので、酵母細胞内のヌクレオチドプールが消費された時点で、S期の進行が停止する。この細胞からDNAファイバーを調製し、取り込まれたBrdUを蛍光抗体で検出するという方法で、検出された初期開始領域は平均6.9kbであった。アルカリショ糖密度勾配遠心で複製鎖を分離した結果、BrdUの取り込みはファイバーで検出されたサイズと同等のサイズの断片にのみ検出され、ファイバーで検出されたものが初期複製開始点を含むことを確認した。FISH法を利用して特定領域の複製開始部位を検討しつつある。BrdU標識されたフラグメントを濃縮し、DNAマイクロアレイに供し、初期複製開始点を網羅的にマップしたい。
2.動物細胞ゲノムのレプリコン解析法を開発した。付着系細胞にビオチン、及びジゴキシゲニン標識ヌクレオチドをビーズローディング法で導入した細胞からDNAファイバーを作製し、複製部位を蛍光検出すると、複製フォークの進行と近接する複製開始部位の存在が可視化できた。この方法により、真核細胞ゲノムのレプリコン構成に関するモデルを提示した。また、マウスメジャーサテライトDNA領域内の複製開始部位の検出に成功した。ゲノム上の特定部位の複製開始点をFISH法により解析中である。
