酵母染色体組換えにおける染色体機能装置の連携

研究概要	本年は最終年度として以下の成果を得て全体のとりまとめを行った。 1.分裂酵母の減数分裂期組換えホットスポットM26におけるクロマチン再編成・DNA切断に関わる因子として、新たにATP依存性クロマチン再編成因子Snf22(特許出願中)を同定した。この遺伝子を破壊すると、M26におけるクロマチン再編成と組換えホットスポット活性が消失した(Yamadaら、EMBO J印刷中)。また、ストレス遺伝子の転写活性化においても同様なメカニズムのクロマチン再編成が重要な役割を果たしていること(Hirotaら、NAR 2004)、転写抑制因子であるTup11とTup12がM26におけるクロマチン再編成の抑制を行っていること(Hirotaら、Genetics 2003)、天然のM26様部位における減数分裂期組換え開始シグナルであるDNA切断の存在、などを明らかにした。これまでの結果から、分裂酵母M26ホットスポット活性化の分子機構の大筋を明らかにした。 2.出芽酵母の組換え開始に関わるMre11とSpo11の減数分裂期染色体における分布を、白髭班と共同で高密度ゲノムタイリングアレイを用いて解析した。その結果、Spo11がまずセントロメア領域に結合してからDNA複製に応じて染色体両腕部に再配置すること、Mre11がDNA複製を機にSpo11と同様の局在を示すようになること、DNA切断が導入される時期になると、減数分裂期特異的コヒーシンRec8の分布箇所(相同染色体のコア部分)の両側(ループ部分に相当)に配置されるようになること、などが初めて明らかになった。以上の結果は、これまで不明であった組換え開始のメカニズムにおいて重要な知見を与える極めて貴重な成果である。
