研究概要	本年度得られた主な研究成果は、以下の通りである。 (1).膜型MMP(MT1-MMP)のがん細胞浸潤・転移での役割:ヒト甲状腺癌、口腔癌、グリオーマでの解析から、MT1-MMPによるproMMP-2活性化がこれらのがん細胞浸潤・転移に関わることを明らかにし、MMP-2のがん組織内での活性をin situ zymography法の開発により初めて証明した。 (2).細胞膜上で働くMMP-7の機能解析:ヒト大腸癌、子宮内膜癌、肺癌などで転移と相関するMMP-7に関して、proMMP-7をbaitにしたyeast two hybrid法でヒト肺組織cDNA libraryをスクリーニングし、テトラスパニンの一分子であるCD151をクローニングした。Two hybrid assayや免疫沈降のデータから、proMMP-7のプロペプチドが本分子の細胞外ドメインと結合することを決定した。 (3).MMPによるVEGF(Vascular endothelial growth factor)マトリクライン作用:血管新生因子であるVEGFが軟骨組織で発現されることを示すとともに、VEGFの血管新生誘導活性がCTGF(Connective tissue growth factor)のスロンボスポンジンモチーフとの結合によりブロックされることを明らかにした。 (4).ADAMTS4の酵素活性阻害実験:ADAMTS4の酵素活性はMMPの組織インヒビター(Tissue inhibitors of metalloproteinases=TIMP-1,2,3,4)のうち、TIMP-3で特異的に阻害されることを証明した。
