研究概要

本年度得られた主な研究成果は、以下の通りである。
(1).MMP-7(マトリライシン)の機能解析:潜在型MMP-7の結合分子を探索するために、潜在型MMP-7をbaitにした酵母のtwo-hybrid systemで肺cDNAライブラリーをスクリーニングしCD151をクローニングした。Two-hybrid assay、免疫沈降、結合実験のデータから、潜在型MMP-7のプロペプチドがCD151の細胞外ドメインと結合することを決定した。さらに、その結合により潜在型MMP-7が活性化されることを実証した。これらのことから、ヒトがん細胞の浸潤・転移に関与するMMP-7にも、潜在型MMP-2/MT1-MMPのような細胞膜表面での活性化機構が存在する可能性を明らかした。
(2).MMPによるVEGF(Vascular endothelial growth factor)マトリクライン作用:血管新生因子であるVEGFが無血管組織である軟骨組織で発現されることを示すとともに、VEGFの血管新生誘導活性がCTGF(Connective tissue growth factor)のスロンボスポンジンモチーフとの結合によりブロックされることを明らかにした。さらに、MMPはVEGF/CTGF複合体のCTGFを特異的に分解し、VEGFの血管新生活性の再出現を証明した。以上の結果からMMPがCTGFの分解を介してVEGFのマトリクラインに関わることを示した。
(3).膜型MMP(MT1-MMP)とMMP-7のヒトがん細胞浸潤・転移での役割:種々のヒト癌組織とグリオーマでの解析から、MT1-MMPによる潜在型MMP-2活性化と、CD151を介した潜在型MMP-7の活性化ががん細胞浸潤・転移に関わることを示した。
