植物細胞におけるアクチン系細胞骨格構築の分子機構

研究概要	植物細胞内において、アクチン系細胞骨格が重要な役割を果たしていることが明らかになりつつある。本研究においては、アクチン系細胞骨格の植物内における機能発現に関して、生理学的、生化学的および形態学的な解析を行い、以下のような成果が得られれた。 1.アクチンフィラメントの束化におけるビリンの役割:植物細胞内において、アクチンフィラメントは多くの場合、束を形成している。しかしながらその分子機構については明らかではなかった。我々は植物細胞内においてアクチンフィラメントを束化するビリンの単離に成功し、その機能がカルシウムとカルモジュリンによって制御されることを明らかにした。 2.トチカガミの根毛において、原形質糸の構築にはアクチンフィラメントが主体的な役割を果たしている。アクチンフィラメントの束による細胞内構造の維持にはタンパク質リン酸化が重要な役割を果たしていることを明らかにした。 3.植物細胞においてミオシンがいろいろな生理的機能に関与していることが明らかになりつつある。しかしながらその特異的な阻害剤は知られていなかった。我々はbutanedione monoximeが植物ミオシンの活性を阻害することを初めて明らかにした 4.植物細胞内において、アクチンフィラメントの構築が微小管によって制御されていることが知られている。しかしながら、その分子的な実体は明らかではなかった。我々はタバコ培養細胞からアクチンフィラメントと微小管を結合させるタンパク質を単離し、そのアミノ酸配列の決定に成功した。。
