魚類の温度適応分子機構の解明と利用

研究概要

魚類は変温動物で、その体温は水温の変化に応じて大きく変化するが、その生体成分を包括的かつダイナミックに変化させて体内の恒常性を保ち、生命活動を維持する。本研究は、温度情報がホルモンを介してどのような機構で遺伝子およびその発現調節まで至り、細胞小器官まで含めた魚類の代謝調節に結びつくのかを総合的に調べることを目的とした。
まず、霞ヶ浦産のコイ科魚ワタカ、ニゴイ、ゲンゴロウブナ、ハクレン、コイ科以外のオオクチバスおよぴブルーギル(サンフィッシュ科)につき、冬季の低水温時に普通筋筋原線維Mg^<2+>-ATPase活性は高く、夏季の高水温時に低いことを示した。しかしボラ(ボラ科)では、夏季と冬季に明瞭なATPase活性の差違はみられなかった。
次に、コイ、タイ、ニジマスを対象に、JC-1試薬で蛍光強度とミトコンドリアの膜電位の相関を、K^+に選択的な中性イオノフォアのvalinomycinを用いて調べたところ、細胞外K^+濃度を上昇させるにつれ、ミトコンドリアの膜電位は減弱した。この結果を、Nernstの式を用いて較正をしたところ、両者には一次関数的な関係がみられ、抽出を必要としない、経時的な変化を追うことのできる代謝活性評価法が確立された。
さらに、メダカは温度馴化に伴ってMg^<2+>-ATPase活性の異なるミオシン重鎖を発現すること、その発現が転写レベルで調節されていることを示した。また、ミオシン重鎖遺伝子を全て含むBACクローンを選び、約295kbpの全塩基配列を決定し、ミオシン重鎖遺伝子の温度依存的な発現には5'上流調節領域が重要であることを明らかにした。
