歯周病におけるプロテアーゼ機能と生体反応に関する基礎的研究

研究概要	歯周病は歯槽骨を含む歯周組織の広範囲な破壊を特徴としており、宿主細胞ならびに口腔細胞由来のプロテアーゼの関与が考えられる。これまでの筆者らの研究から、歯肉縁下プラーク中に存在するグラム陰性嫌気性細菌Porphyromonas gingivalisが産生する2つの主要なシステインプロテアーゼ[Arg-gingipain (RGP)とLys-gingipain(KGP)] と宿主細胞由来のリソソーム性プロテアーゼ(cathepsin B,cathepsin L,cathepsin G,medullasin)の本疾患への関与を示唆してきた。とくに興味深いことには、P.gingivalisが産生するRGPおよびKGPは宿主に対しては強力な破壊因子として働いて病原性を発揮するが、菌体自身には病源性を示すことはなく、むしろ有用であるということである。本年度の研究は、RGPおよびKGPがこうした機能を発現し得る分子メカニズムを知る目的で行われた。そのために、RGP遺伝子(rgpA rgpB)およびKGP遺伝子(kgp)をそれぞれ完全に欠損させた変異株と両酵素遺伝子を同時に欠損させた三重変異株を作製し、それらの性状を野生株と比較しながら詳細に解析した。各変異株の培養上清を用いた実験では、RGPおよびKGP欠損株は野生株のもつヒトタイプIコラーゲンや免疫グロブリンに対する分解能を有意に減少させるが、依然として強い分解能を保持していた。しかし、両酵素を同時に欠損させた三重変異株では、これらの蛋白分子に対する分解能は完全に消失していた。野生株のもつ好中球に対する強い傷害活性も、RGPおよびKGPの単一酵素欠損株では有意に抑制されるものの依然として傷害活性は残存しているが、三重変異株では完全に消失した。これらの結果は、菌体外へ産生分泌されたRGPおよびKGPが、歯周組織の主要蛋白質の分解や生体防御系の破壊を惹き起こして、歯周病の発症や進行に深く関わっていることと強く示唆されている。各変異株と野生株を各種アミノ酸を含む通常のBHI培地で培養した時、いずれの変異株も野生株と同様の正常な増殖カーブを示した。しかし、ウシ血清アルブミンのみを栄養源とする合成培地で培養した時には、単一酵素欠損株は野生株と同様に増殖するが、三重変異株はほとんど増殖できなかった。これらの結果は、本菌の発育増殖に必須のアミノ酸・ペプチドの獲得はRGPおよびKGPの両酵素によって行われていることを示している。
