脊椎動物咀嚼器形態の発生メカニズムと進化

研究概要	すでに前年度記述したように、顎口類において顎骨弓の形態パターニングは、表皮に発現する成長因子と、その作用によって制御される、神経堤間葉での局所的なホメオボックス遺伝子発現に負うところが大きい。これは我々の顎を形作る最も基本的な発生プログラムのひとつであり、これら遺伝子の発現パターンが形態形成の基本方針を教えてくれるとも言うことができる。そこで、直ちに我々はヤツメウナギにおけるFgf8,Bmp4と、それらの標的と目されるMsx, Dlx遺伝子の相同物の単離に踏み切ったわけであったが、今年度はそれらすべての単離にようやく成功し、その発現パターンをも解析することができた。驚くべきことに、それは、ヤツメウナギの上下の唇と、我々の顎が相同だという、従来の見解と食い違う結果をもたらした。この論文は、今年度印刷されたが、それは、純粋に形態学的パターンに基づく比較を行うと、ヤツメウナギと顎口類では、口周囲の間葉の用い方が全く異なっており、したがって顎と上下唇は別物である、というものであった。遺伝子発現と形態要素。これらのどちらか一方の相同性を認めると、他方が食い違う、という状況が生まれたわけである。この不一致は、おそらく表皮に発現する成長因子の広がりが、進化的に変更したためではないかと推測し、ニワトリ胚を用いて局所的にFGF8リコンビナントビーズを移植することで、標的遺伝子の発現パターンを確かにヤツメウナギの発生過程のパターンに類似させることができた。つまり、進化的意味における表現型の模写を作出する事ができたわけである。顎の進化はヘテロトピー、すなわち組織間相互作用の位置のシフトを基盤とするというこの結論をまとめた論文は、現在投稿中の段階にある。
