酵素による糖質の炭素環化を基軸とする持続可能な有機化学工業資源の開拓

研究概要

(1)Bacillus circulans由来2-デオキシーscyllo-イノソース(DOI)合成酵素遺伝子(btrC)の大腸菌発現系を用いて糖質環化生成物DOIの生産条件を、培地、グルコース量、酵素誘導とグルコース添加の時期等を対象に、既報のDOI定量法を用いて検討し、グルコースから25%程度変換出来る条件を見い出した。
(2)糖質と類似した性質を示すDOIを培養液中から分離する条件を検討し、先ず活性炭による吸脱着で來雑物を除去し、その後陽イオン交換樹脂及び合成吸着剤にて精製する方法を確立した。
(3)DOI合成酵素遺伝子(btrC)の大腸菌以外の微生物による発現を検討した。先ず酵母への組み換えを検討し、その発現に成功した。ウェスタンブロツトでタンパクは確認できたが、酵素活性が検出できなかった。同様に枯草菌での発現を目指し、発現用プラスミドを構築した。
(4)抗生物質生合成系の遮断によるDOI生産を目的として、最近発見したDOIへのアミノ基転移酵素BtrSの遺伝子破壊を試みた。btrSの部分欠失遺伝子を構築し、既に確立した方法で抗生物質生産菌B. circulansに導入した。現在相同的組み換えを行って破壊株を検索中である。
(5)他種微生物由来の糖質環化酵素を新たに探索すべく、カナマイシン生産菌Streptomyces kanamyceticusゲノムについてbtrS類似遺伝子を指標に検索して、btrSに対応するkanSとともに、btrCに対応するDOI合成酵素遺伝子kanCを放線菌で初めて同定した。
(6)有機工業資源としての適用拡大に向けたカルバグルコースヘの誘導を検討した。カルバグルコースは甘味を持ちながら糖質の代替化合物となりうるため、生活習慣病対策のひとつとして期待されている。培養液から抽出したDOIから6工程でこれを合成する簡便な方法を開発した。
