研究概要	(1)SLEの特徴的病態の一つに、B細胞の異常活性化に伴う自己抗体を含む高IgG血症があげられる。この現象も明らかに遺伝的素因の関与する現象である。SLE自然発症モデルマウス系を用いて、ゲノムワイドにB細胞の異常活性化の感受性遺伝子を解析したところ、その一つが第1染色体テロメア側にmapされた。このごく近傍にB細胞活性化抑制分子であるFcγRIIB遺伝子が存在したので、FcγRIIB遺伝子の塩基配列をSLEマウスで解析した結果、転写制御領域を含むプロモーター領域に2カ所と第3イントロンに1カ所の塩基配列欠損が認められた。この多型は遺伝学的にB細胞異常活性化に良く相関しており、SLE主要感受性遺伝子の一つである可能性が示唆された。 (2)SLE合併抗リン脂質抗体症候群(APS)のモデルNZWxBXSB F1雄マウスに生ずるAPS三症候、抗カルヂオリピン抗体、血小板結合抗体、血小板減少症に寄与するBXSB感受性遺伝子の遺伝子座mappingをNZWx(NZWxBXSB)F1退交配マウスを用いたマイクロサテライトDNA解析で行った結果、各症状に共通する主要感受性遺伝子が染色体8と17にmapされた。このうち第8染色体遺伝子座領域には、有力な候補感受性遺伝子として組織plasminogen activator遺伝子(Plat)が、また、第17染色体遺伝子座領域にはpalsminogen遺伝子(Plg)やMHCが存在したので、各々の遺伝子の塩基配列を調べたところ、Platのcatalytic domainにBXSB特有のアミノサン転換を伴う単一塩基多型が発見された。この領域はplasminogenをplasminに変換する酵素活性を示す部位であり、この多型が有力なAPS感受性遺伝子である可能性が示された。
