研究概要

本研究は、咀嚼、嚥下等の口腔機能の加齢に伴う変化が、いかなる中枢神経系の変化によってもたらされるかを知ることにより、口腔の健康維持に役立てようとするものである。また、加齢に伴う口腔機能の低下が全身にもたらす影響を調べることにより、口腔の健康の重要性を明らかにしようとするものである。本年度は、後者について研究を進めた。加齢に伴い口腔あるいは全身的な疾患により、軟らかい食品しか摂食しえない場合を想定し、実験的に軟食を続けた場合と軟食を続けてから、硬い食品を与えた場合に痛みに対する感受性に変化が認められるかどうかについて調べた。雄性SD系ラットを用い、以下の2群に分けて、実験を行なった。ひとつは軟食群で、軟食を10日間与え、起炎物質であるCFAを足底皮下に注入し、その後も軟食を与え続けて3日後に灌流した群、もうひとつは硬食群で軟食を10日間与えCFAを足底皮下に注入し、1日後から硬食を与え3日後に灌流した群である。熱刺激に対する逃避潜時からCFAを足底皮下に注入後にみられる疼痛閾値低下は、軟食群に比べ硬食群で有意に抑制された。L4、5の脊髄後角部に発現する疼痛情報の指標であるc-Fos陽性細胞は軟食群で注入側に多数認められた。これに対し、硬食群では少数認められたのみであった。また、ナロキソンの持続注入により硬食群で観察されたc-Fos陽性細胞発現の抑制は消失した。以上のことから、軟食摂取から硬食摂取に変えた時にみられる疼痛抑制は、オピオイドを介して発現する可能性が示された。
