研究概要	本研究では、プロスタグランジンの脳機能、特に神経細胞のアポトーシス阻害作用に焦点を絞り、プロスタサイクリン(PGI_2)関連化合物の脳内動態および標的受容体の機能の解明とその抑制を目指して研究を推進し、いくつかの成果を上げることができた。まず、脳内PGI_2受容体探索分子15R-TICの陽電子放射断層画像撮影法(PET法)への適用のため、パラジウム触媒を用いた高速メチル化反応を開発し、PETトレーサー合成条件下での合成プロトコールを確立した。また、15R-TICの主な作用として本研究者らが発見した高酸素状態で引き起こされる神経細胞アポトーシスの阻害作用について、15R-TICの誘導体を設計・合成し、構造活性相関研究を行った。さらに、得られた知見をもとに15R-TICの標的であるPGI_2脳内受容体の構造と生理機能解析のための新規リガンドの設計・合成に取り組み、有望な誘導体の創製に成功した。続いて、探索分子の放射核標識体を合成してラット脳凍結断片のオートラジオグラフィーを行い、中枢型IP_2受容体の局所分布の視覚化に成功するとともに、探索分子の動態解析を行い、IP_2が情動行動、嗅覚、記憶、学習、痛覚、聴覚を掌る部位に局在することを明らかにした。一方で、TICおよび関連化合物の生物活性、特に、in vivo海馬ニューロンのアポトーシス阻害効果とin vivoラット脳虚血モデルでの作用評価のためのモデルの構築にも成功した。また、IP_2受容体タンパク質捕獲のための新たな光親和性標識リガンドを設計し、その放射核導入体の合成に成功した。現在、光親和性標識実験を検討している。以上、本年度の研究は概ね計画通りに遂行できた。
