FRETによるニューロンの転写因子間相互作用の可視化

研究概要	平成11年度に行った研究は、まずFRETの基礎的な問題点の整理を洗い出し、どのような蛍光物質をどのような形でニューロンに導入するのが最も高感度で特異性があるのか検討を加えた。現在使用可能なものとしては、GFPの改変型であるCFP(cyan fluroscent protein、励起波長433/453nm、蛍光波長475/501nm)とYFP(yellow fluroscent protein、励起波長513nm、蛍光波長527nm)が挙げられる。それぞれの蛍光色素に異なる転写因子(低分子脂溶性物質レセプター)を遺伝子的に融合させ、培養細胞(COS細胞、初代神経系細胞)へこれらの遺伝子を共発現させた。グルココルチコイドレセプター(GR)にはCFPを、ミネラルコルチコイドレセプター(MR)にはYEPを結合させた。これらの状態においてGRやMRに対するアゴニストやアンタゴニストを投与し、細胞内での蛍光特性の変化をリアルタイムイメージング法によって観察し、異なる転写因子が同一細胞でどのような立体構築的相関性を有しているのか検索した。高濃度のコルチコステロン投与実験ではCFP-GRとYFP-MRの動態に著しい差異は認められなかったが、低濃度のコルチコステロン投与実験においてはYFP-MRの細胞質から核への移行がCFP-GRに比べて時間的にはやく、濃度による変化が観察された。また、リガンドに対する特異的細胞内動態も観察され、この系が実験系として充分な妥当性を有していることも確認できた。
