FRETによるニューロンの転写因子間相互作用の可視化

研究概要	平成12年度においては、GFPの改変型であるCFP(cyan fluroscent protein、励起波長433/453nm、蛍光波長475/501nm)とYFP(yellow fluroscent protein、励起波長513nm、蛍光波長527nm)を用いて、各種核内レセプターについて検索した。glucocorticoid receptor(GR)やmineralocorticoid receptor(MR)、androgen receptor(AR)はリガンドと作用しない場合は通常細胞質内に存在していたのに対して、estrogen receptor(ER)a/b、progesterone receptor(PR)は核に分布していた。また、共役因子であるSRC-1やp300などはその機能からも核に分布していることが容易に予想されるが、これらのことを培養細胞による遺伝子発現系で確かめた。FRETはそのバックグラウンドをいかに処理するかに正否がかかっており、リガンドが作用しなくても核に存在する(ER)a/bと共役因子をCFPとYFPで標識し、リガンド添加によって核内でのFRET値を算出した。FRET値は供与体と授与体の蛍光強度から授与体の蛍光強度を差し引き、その値を授与体の値で割ったものとなるとした。FRET値が上昇するものは核内で一定の領域に留まるものと、さまざまな領域に変移するものに分けられた。これらは核内でのそれぞれの働きを動的にしめしていると考えられた。今後は、細胞質から核への移行に伴うFRETについて研究する必要性が生じた。
