機能電極界面を用いた金属タンパク質の電子移動反応速度制御因子の解明と機能解析

研究概要

本研究では、原子・分子レベルで制御された機能界面を用いて金属タンパク質分子、特に、ミオグロビンおよびフェレドキシン、さらにその改変・変異分子を用いて、その電子移動反応の詳細を明らかにすることを目的としている。本年度は、以下のような研究成果を得た。
1.金属タンパク質の抽出・精製について、現在、市販されていない金属タンパク質、特に熱安定性に優れたクロレラ由来のフェレドキシンを抽出・精製する手法を確立した。
2.金属タンパク質の電子移動のための単結晶機能界面の創製と表面構造の原子・分子レベルでの評価に関して、フレームアニーリング法および真空蒸着法で作製した平滑で原子レベルで制御した表面上に電子移動プロモーター分子を配向させて、その表面の原子解像のプローブ顕微鏡観察および表面増強ラマンおよび赤外分光法を用いて構造解析し、2-、4-ピリジンチオール(2-PySH、4-PySH)、2-ピリミジンチオール(2-PymSH)等の界面構造や吸着分子のピリジン窒素のpKaは変化などを明らかにした。4-PySHの金(100)面上での吸着構造やアントラキノン部位を持つアルキルチオールの金電極上での酸化還元に伴う吸着構造の変化を明らかにした。2-PymSH修飾金電極上では銅タンパク質であるアズリンの速い電子移動反応が観測できることを明らかにした。
3.ミオグロビン及び変異分子の電子移動反応速度制御因子の解明に関して、ミオグロビン、ヘム部位を改変した半人工ミオグロビン、およびアミノ酸変異分子を用いて、電極上での電子移動速度とミオグロビン構造との相関を明らかにした。
4.金属タンパク質固定化素子の構築と電子移動反応機構の解明について、金単結晶電極面上に種々の方法で金属タンパク質を特定の配向で固定化できることを明らかにした。
