炭素・窒素化合物とその輸送体のジオグラフィック・マッピング

研究概要	In situ技術を植物に応用し、作物特にイネの生育・生産性の重要要因である炭素・窒素化合物の代謝・輸送機構についての全く新しい視点の導入、かつそれに付随する各種技術・実験手法の確立を達成目的とする。炭素・窒素化合物の輸送・変換解析: 炭素放射性化合物を植物体(最初にイネ培養細胞を用いた)に導入し、その代謝中間体と輸送過程について経時的に解析した。糖飢餓を施した細胞では糖の取り込み活性の上昇が観察された。これがトランスポーターに由来するものかについて検討中である。炭素・窒素輸送体(トランスポーター)のクローニング: 炭素化合物輸送体として、スクローストランスポーター(OsSUT1)と単糖トランスポーター(OsMST1-5)のcDNAとゲノムクローニングを行った。In situ mRNA検出の結果、OsSUT1は維管束篩部伴細胞で、またOsMST3は根の内表皮細胞と葉鞘のsclenchyma細胞・肥厚細胞で発現していることを明らかにした。窒素輸送体としては、イネにおいてアンモニアトランスポーター(OsAMT1-3)と硝酸トランスポーター(OsNIT1-2)のcDNAクローニングを行った。現在それらの輸送機能と細胞(特に根における)発現調節について解析している。
