研究概要	1.Cucumis figareiから単離したCF-RIP遺伝子の解析、遺伝子産物の発現、遺伝子導入:ゲノムDNA由来のCF-RIP遺伝子と、mRNA由来のCF-RIP遺伝子のの相同性は100%で、ORF中にイントロンは存在しないことが判明した。また、大腸菌によるCF-RIPの発現は、pET15-bへCF-RIP遺伝子を挿入し、BL21(DE3)pLysSで発現させた場合にのみ認められた。可溶化後のCF-RIPには抗ウイルスおよび抗Pythium活性が認められ、現在RIP活性を確認するとともに抗体を作成中である。さらに、遺伝子導入実験ではタバコ培養細胞(BY-2)でのCF-RIP遺伝子の発現が確認された。現在、35SCaMVプロモーター、PRla遺伝子プロモーター、およびC.figa reiより単離したCF-RIP遺伝子プロモーターを連結したベクターを構築し、アグロバクテリウム法によりタバコに導入し形質転換体を作出中である。前者については当代でのCF-RIP遺伝子の発現が確認できた。 2.Cucumis figareiから単離した抗ウイルス活性物質フイガレンの機能解析と作用機構:クローニングされた3'側581塩基のフイガレンcDNA断片の塩基配列を決定した。ホモロジー検索の結果、相同性の認められた蛋白質について、各々の活性を調べたところ、フイガレンにはRnase活性が認められたが、核酸結合能は認められなかった。フイガレンの葉面処理による抗ウイルス活性は感染阻害と移行阻害によることが判明した。FISH法を用いた蛍光顕微鏡観察では、局部感染宿主の処理葉ではその多くはサブリミナル感染で、全身感染宿主では、特に感染初期の感染細胞から健全細胞へのウイルス移行を阻害することが明らかになった。なお、処理部位以外でも阻害作用が認められ、フイガレンの浸透移行によるか、あるいは何らかのシグナル物質の移行により阻害作用が発現しているものと考えられた。
