イネにおける葉の光合成の生化学は個体成長と乾物生産をどこまで説明できるか?

研究概要

イネを実験材料に光合成産物の成長への有効利用率について呼吸ロスによる評価を行った。具体的には夜温のみを変化させた条件で栽培を行なった。昼温/夜温として27/27℃区、27/22℃区、および27/17℃区を設定し、植物の個体レベルでの成長解析を行なった。相対成長率(RGR, relative growth rate)、NAR、LAR等について調べたところ、標準区、各夜温処理区でのイネの成長の特徴に余り大きな差はなかった。ただし、27/17℃区において若干乾物量が少なく、呼吸は必ずしもロスになってないことが示唆された。夜温の低下はLARの低下に影響を及ぼし、そのことが若干の乾物量低下の要因であった。また、夜温の低下はデンプンの蓄積量低下につながったが、それは成長様式や個体としてのバイオマスに影響を与える要因にはならなかった。その他、新しい実験材料としてrbcS sense植物の作出を試みた。以前、遺伝子の構築は、Anti-rbcSイネを作出に用いたプラスミ・ドpRASとrbcScDNAをsense方向に組み込んだpRS(未発表)よりrbcS promoterとrbcScDNA部位を切り出しアグロバクテリウムの形質転換用ベクターpBI101Hmにセンス方向とアンチセンス方向に組み込むことによって行った。構築したプラスミッドをアグロバクテリウムに形質転換させた後、イネカルスに感染させ、Sense-rbcSイネの再分化を試みている。
